The National Roads Agency said an air ambulance was called to a “serious” motorway accident involving multiple vehicles.
The southbound carriageway of the A1(M) is closed between junction 16 (towards Stilton, near Peterborough) and junction 13 (towards Brampton, Cambridgeshire).
Earlier, northbound carriageways were blocked to traffic to allow the air ambulance to land, but were later reopened.
Police and the East of England Ambulance Service also attended the scene.
The National Highways Agency said: “All emergency services remain at the scene, with Cambridgeshire Police leading the rescue effort.
“Traffic officers and contractors have implemented strategic closures as the road is likely to remain closed throughout the evening.”
The department has implemented diversions and asked southbound drivers to follow the solid triangle symbols on road signs.
